The formation of global bacteria replication Brucella is the key to survival in intracellular and cell proliferation in host cells. Research has found that Brucella can use host cells of small G protein Sar1 form replication of bacteria bubble, and Brucella type IV secretion system can also regulates replication of bacterial vesicle formation, but Sar1 and type IV secretion system between the relationship is not clear. Our pre experiments show that Sar1 can be combined with effector proteins. Accordingly, we hypothesized that Brucella by type IV secretion system to secrete effector proteins get Sar1 host cells to regulate the replication of bacterial vesicle formation. In order to confirm this hypothesis, this paper intends to the GST -Sar1 fusion protein settlement, two-dimensional electrophoresis, mass spectrometry, yeast growth inhibitory effect experiment and protein expression assay to screen the type IV secretion system and effector proteins capable of interacting with the Sar1; and then construct the lack of effector proteins of Brucella strains; by mid detection of Brucella bacteria vesicles and lysosomes and endoplasmic reticulum fusion to explore the effect of protein in replication bacteria of bubble formation in the role. The implementation of the project for Brucella cell survival mechanism of new ideas, and found a key effector proteins may provide potential targets for the treatment of brucellosis。
